Parmi les 170 modifications, le réalésage de 4 mm permettant au très beau moteur 6 cylindres (signé Tadek Marek) de passer à une cylindrée de 4L pour développer une puissance de 285 chevaux, une nouvelle boîte manuelle synchronisée ZF 5 vitesses (à partir de 1964) et trois carburateurs Weber 45 DCOE.
Ce qui contribua à la notoriété internationale de la DB5, c'est évidemment son apparition dans " Goldfinger " où elle devient la voiture de James Bond, le plus célèbre des agents secrets.
